If you are doing proteome-wide untargeted experiments then we would recommend that your computer have at least 64GB RAM because Skyline wants to keep data about all the peptides in memory at the same time.

If you are doing targeted experiments you could get away with having much less memory in the computer.

Skyline requires 64 bit Windows with an Intel, AMD, or other AMD64 CPU.
