How to do normalization by 3 heavy compound for 3 more compounds xue shi  2021-09-14 10:26

Dear Skyline team,
I know how to do normalization just by one compound normalized by one heavy compound by skyline. However, Do skyline have the ability to normalize 17 compounds by 3 heavy compounds? Not every compound has heavy compound as internal standard. If yes, How should we process this in transition list? I attached my transition list in the attachment.
I really appreciate if you could help with this.
Thank you,
Katie

Skyline lets you specify that a particular molecule should be normalized by doing the ratio to a different molecule in the document.
This feature is called "Surrogate Standards".

There is a description of how to use this feature here:
https://skyline.ms/wiki/home/software/Skyline/page.view?name=Surrogate%20Standards

After you have imported your Transition List into Skyline, you can right click on Acetic, But, and Prop in the targets tree and choose "Set Standard Type > Surrogate Standard".
And then you can use the Document Grid to change the Normalization Method of your other molecules to the appropriate value ("Ratio to surrogate Acetic (heavy)" etc.).
