Quantifying with surrogate standards Joerg  2020-11-24 09:10

Hi,
I am tring to quantify different lipid classes using several surrogate standards. Although I set the respecitve molecules as "surrogate standard" and set the "Normalization Method" to the respective standards for the other molecules, there is no option to normalize the area view to these standards. Nor is it possibl to use the surrogate standards in the Quantification tab of the "Molecule Settings". I am obviously missing something...
Many thanks in advance!
Joerg

Using that normalization method in the Area graph is a new feature that will be available in the next update of Skyline-Daily (which will happen in a few weeks).
For now, the only place to see that normalized value is in the Document Grid. In the Report Editor, the column which shows you the normalized area is:
Molecule Groups > Molecules > Molecule Results > Quantification > Normalized Area

This is exactly what I want to do. Once I set a group of transitions as surrogate standards how do I integrate the rest of the peaks based on these surrogate standards?
Thanks

Once you have specified which molecules are Surrogate Standards, you need to create a report in the Document Grid that includes the "Normalization Method" column and then you can change the Normalization Method of the molecules that you are interested in to "Ratio to Surrogate Xxx".

Here is a page that talks about how to use Surrogate Standards:
