Hi,
I have skyline 20.2 64 bit on my Windows 10. To get familiar with skyline and ion mobility data , l followed the tutorial 'Skyline Ion Mobility Spectrum Filtering' and I downloaded zip file. I read that with new version of skyline IMS filtering settings moved to Transition Settings - Ion Mobility. My problems is: 'Use Results' button is not clickable. Why?

You'll need to use the Create button to create the library before you can Use Results to populate it.

Thank you very much for the quick reply.

I create a library, but cliking on the apex of the yellow chromatogram I can't see the horizontal violet range showing the filter imposed.
For my analyses, I am using skyline for analysis of peptide library, analysed with HDMSe. I import FASTA file and then results ( raw file).
It's correct?
I am interested in understanding Ion Mobility Separation.

You just need to reimport the raw file (Edit > Manage Results > Re-import) so that Skyline can use the newly created ion mobility library for improved chromatogram extraction.

I followed your instructions, but clicking on the apex of the y6 chromatogram to show the Full-Scan graph, the horizontal violet range showing the filter imposed by the drift time predictor is not present.
In this case it means that the filter has not been applied?
I have attached the screenshot

Hi Mariangela,

It would probably be most efficient if I could have a look at your Skyline document (save it with File > Share > Complete, this creates a .sky.zip file that also includes the library) and the raw files that are giving you trouble. You can upload to http://skyline.ms/files.url or we can arrange something private if you wish.

OK, with your files in hand I find that simply doing the reimport works just as expected.

If you find that this is not the case for you, we can investigate further, but I think you just missed a step.
