Adjusting a few RTs james.broadbent  2017-06-19 04:57

Dear Skyline Team,

Can I quickly check - is there any way to adjust a few RTs in a spectral library without adjusting all RTs? I am working with a lab that have run some samples and more recently some heavy standards and we wish to line up the heavy and light peptides manually.

Kind regards,

James

Hi James,

Before I make any suggestions, I'd like to be sure I understand the scenario. You're wanting to add the heavy peptides to an existing light-only spectral library? How was the original library generated?

Thanks
Brian

Hi Brian,

Sorry for the delay. I'm wanting to add some additional heavy peptides to a library, but these IDA data were run matrix-free and I can see an RT shift based on the light versions. The original library was created from an IDA run of light biomass spiked with heavy proteins. There is good coverage for most heavy proteins but a few only have light peptides. I am working to edit the group.xml before import but the xml import is causing an error. Kaipo is helping with the xml import.

Kind regards,

James
