Hi Brendan,
 
we have a project where we are using enzyme combinations.
 
It is easy to define enzyme combinations if say both enzymes are C-terminal or both enzymes are N-terminal with their cleavage specificity. However, here we have a combination where one is N-terminal and one is C-terminal.
 
How do we define that in Skyline ?
 
See screenshot, that is what we would like to combine (and then we have more examples just like that).

in Skyline can we allow combinations of enzymes, or custom enzymes with AAs for both directions.

This was fixed for v1.4. It is now possible to define an enzyme (protease) that cleaves both C-terminal to some AA residues and N-terminal to others.
