Hello, Thank you very much for this wonderful program. I am trying to add a heavy 13C amino acid residue to a modified peptide but I haven't been able to do it. I've tried the settings tab --> peptides settings and I have also tried the edit --> refine --> advance --> add, but I don't see the I (C-term) bold or modified (+6). After adding the heavy 13C in settings, I go to the specific peptide do RMC --> Modify --> edit modifications, I can't see the heavy1 modification in the Isoleucine drop down list, created previously in settings.

Please post a screenshot of your settings and what you are seeing in RMC > Modify.

Hello Brendan, thank you very much for your help and for the link. I glanced at the tutorial and I may be able to get to the heavy residue added to the peptide sequence. I am sending the settings and the RMC --> modify. In the transition settings I have 2,3,4 precursor charges and 1,2 ion charges for y ions. Again, thank you very much.

That is strange. I wouldn't expect things to look like what you are showing. I guess I will need to have a look at the document to try to make sense of it.

Can you use File > Share - Complete to create a .sky.zip which you can upload to our file sharing point?
