Export method on Thermo Altis QQQ kristin geddes  2018-11-16 08:30

I just had a Thermo Altis QQQ installed this week in my lab. I am trying to export a Skyline method and I am getting the error "Error: Failure during initialization. TSQ-EZ Method support may not be installed. Method export for a TSQ should be performed on the TSQ instrument Control computer and requires TSQ version 2.3 or better" (see attached screen shot)
I am on the instrument computer and we are running TSQ version 3. Any tips? I have tried both Skyline and Skyline Daily.

Thanks!

Kristin

Hi Kristin,

We have an Altis here in the MacCoss lab so I'll check to see how it's working here. I'll get back to you ASAP.

Thanks for using the Skyline support board!

Here's what our guy had to say:

"Thermo TSQ is for the older model such as Quantum. The introduction of Quantiva make their instrument software not compatible with their old Quantum software. It makes people very confused. I guess for her the workaround is to export Transition List using Thermo Quantiva option. Then import the transition table in the Altis method file."

That's confusing to me - why would TSQ be running on that instrument computer if it's not compatible with that instrument?

It sounds like we might have some work to do to properly support the Altis. But hopefully this gets you moving for the moment.

Hi Kristin,
You want to choose Thermo Quantiva for your instrument method type instead of Thermo TSQ. The Thermo TSQ type is for Thermo QqQ instruments up to and including the Vantage, but starting with the Quantiva and now including the Altis, Thermo switched method writing software. The new software supports method writing for Thermo Quantiva/Altis and Thermo Fusion/Lumos.

Hope this helps. It seems that the researcher doing the most work on the Altis in our lab is simply exporting a Quantiva transition list and importing that into his template method himself manually, rather than using File > Export > Method to have Skyline do that work. Thus a bit of lost knowledge when Jarrett and Kaipo (who teamed up to produce the new method writing implementation for the Thermo Quantiva and Fusion) left the lab in near succession.

Let us know how this works for you. Thanks for the clear screenshot with your post.

Hi Brendan

Thanks for the tip! Unfortunately, that didn't work either. Trying to export using the Thermo Quantiva says that I don't have a registry key and that TSQQuantiva is not installed on this machine. I have attached the screen shot and I get the same message in Skyline and Daily.

Thanks!

Hi Kristin,
I am looking at this. Apparently, the MacCoss lab has been exporting Thermo Quantiva transition lists and importing them into the template methods manually. Seems to work, but I'd obviously like to get back to where you can just Thermo Altis from the instrument list and have it to the right thing for both transitions lists and methods. Looks like it might not be too much work, but we also lost Kaipo who originally wrote the Quantiva method exporter, though it was largely based on other method exporters I wrote.

I will let you know more when I have it, but for now the workaround appears to be Quantiva transition list output and manually importing the transition lists into your template method inside a Thermo method editor.

Hi Brendan

Thank you for the update! I have been successfully doing the export transition list with the Quantiva template and that is working. If there is anything that I can provide to help, please let me know!!

This is fixed in the latest Skyline-daily release (I hope). At least it worked on the MacCoss lab Altis. Let me know whether you are able to use Skyline-daily 4.2.1.18329 for method export on your Altis, and if so, I will merge my changes into the upcoming Skyline 4.2 patch.

Thanks for reporting the issue.

Hi Brendan

I was able to create the method but then when I opened the method file, there were no transitions it in. I have attached a screen shot.

Thanks!
