Job Description:
We are currently seeking a self-motivated, passionate, and accomplished Scientist to join our Analytical Development team. Using your strong attention to detail, you will play a significant role in analytical method development and characterization of therapeutic proteins expressed using SoluPro™, AbSci’s proprietary E. coli expression platform. You will utilize your successful background in the development and analytical characterization of complex biologics by LC-MS and proteomics approaches while exploring new and innovative technologies. This position will utilize your interpersonal and strong written and verbal communication skills for multi-disciplinary collaboration with the other teams within AbSci, including Molecular Sciences, Fermentation, Purification, and Bioinformatics. You bring a passion for scientific innovation, excellent technical skills, and you execute your responsibilities with high attention to detail. You work with a sense of urgency, and thrive in a team dynamic in a high-performance culture.
Responsibilities:
Develop and execute laboratory tasks to support the development and optimization of various mass spectrometry-based methods for the characterization of complex biotherapeutics by LC-MS, including intact mass, disulfide mapping (including PTMs), and targeted mass spectrometry (PRM/MRM)
Perform multi-step sample preparations including enzymatic digestion (solution and in-gel), reduction/alkylation, buffer exchange, deglycosylation, sample concentration, etc.
Method development and optimization for transfer to non-mass spectrometry, stand-alone LC-UV-based techniques for analyses of protein biologics
Operation, maintenance, and installation of LC-MS instrumentation
Write and review formal reports, including method SOPs, reports, and other technical documents
Evaluate data for trends indicative of product performance, method performance, and/or analyst performance and present internally to team members and senior management
Oversee routine instrument operation and maintenance
Lead, mentor, and inspire junior team members
Evaluate new technologies and platforms
Support internal technical teams, including R&D, fermentation and purification process development teams
Maintain accurate documentation of all work in laboratory notebooks
Qualifications:
PhD in Analytical Chemistry, Biochemistry, or related scientific disciplines
Experience in protein engineering, expression, purification, and biochemical characterization (e.g. antibodies, cytokines, and antigens).
Hands-on knowledge of protein analytics and characterization by LC-MS, including intact mass, disulfide mapping, PTMs, and targeted mass spectrometry (MRM/SRM)
Hands-on experience with SDS-PAGE, Western blot, and capillary electrophoresis
Experience in the operation, maintenance, and troubleshooting of associated instrumentation
Strong capabilities in experimental design, execution and data interpretation
Ability to communicate with colleagues and business partners who do not share candidate’s core area of expertise is required
Huge plus (but not required):
Post-doctoral and/or industry experience preferred; 2+ years industry experience is a plus
Experience with Sciex and Thermo MS and Agilent LCs is preferred
Experience with proteomics is preferred
Experience in LC-MS sample preparation for implementation on automated liquid handlers is preferred
Experience with JMP or comparable statistical software packages is preferred
Proficiency in Python or R is a plus
Experience with ELISA, BLI, and SPR is a plus
Experience working with CMO’s and CRO’s for methods transfer, method optimization, and data and report review is a plus
Knowledge of cGLP/cGMP, FDA, EMA, ICH guidance and industry standards for analytical development and characterization of proteins is a plus
