MIDAS format missing taperez  2021-09-21 04:49

Hello,

When I save a file, skyline usually creates and saves a MIDAS format as well. For some reason the files are no longer saving this data format. Is there something Ive accidentally clicked off? Im running Skyline version 21.1.0.146

Thanks!
Tatiana

Hi Tatiana,
Skyline checks for MIDAS spectra automatically when importing results. Specifically, you must have imported MIDAS wiff files (containing spectra) into your document, meaning that other formats like mzML converted from wiff aren't supported.
If you have imported MIDAS wiff files and still are not seeing the expected library, let me know and I can take a look into what's going wrong.

Thanks,
Kaipo

Hey Kaipo,

Yes we import the wiff files with our spectra into skyline but when we look at the saved files as you see below. The midas format is not present. Were not sure why.

Would you mind doing a File > Share and uploading the zip file, along with the MIDAS wiff file(s) to: https://skyline.ms/files.url and I'll have a look?

Thanks,
Kaipo

Hey Kaipo,

Ive input the raw wiff file labeled 20210923_Midas_Issue.zip let me know if you want me to also share the skyline file itself.

Thanks,
Tatiana

Hi Tatiana,
Thank you for uploading the zip file. Was this data acquired differently from ones where Skyline generated a MIDAS library? The files don't seem to contain any spectra, only chromatograms. With the MIDAS workflow Skyline expects to find spectra in the data files as well.

Thanks,
Kaipo

Hey Kaipo,

The acquisition method is the same EXCEPT that the file sent was collected using an unscheduled method. Could this be the reason?

Thanks,
Tatiana

Hi Tatiana,
Sorry, I'm not 100% sure - my guess would be yes but you may want to confirm with Sciex.

Thanks,
Kaipo
